The CAPE Technologies
High Performance Dioxin/Furan Immunoassay Kit is designed
to provide screening (semiquantitative) results at predetermined
or user selected levels. Advanced users may wish to produce
quantitative results in certain situations. Development
of quantitative data using the DF1 kit is possible, but
requires care and experience. The highest probability of
success is for analysis of many samples from one source.
Experience and careful use of quality assurance (QA) samples
will increase the probability of success.
CAPE Technologies
provides the items necessary for quantitative analysis by
EIA. Standards of 2,3,7,8-TCDD in methanol are available
in the DF1-ST
Starter Kit or separately from Wellington
Laboratories. Use of all 4 of these
standards in one EIA run will define the standard curve
described in section D (Table 1) of the kit insert IN-DF1.
The required calculations can be performed using Calculation
Module C, a Microsoft Excel workbook, from CAPE
Technologies.

Because of the intrinsically sigmoid shape
of immunoassay response curves, Module C uses an iterative
non-linear curve fitting procedure based on the same four
parameter equation which is the basis for most commercial
immunoassay data analysis software. Module C calculates
the best fit standard curve and concentrations of unknowns.
